Establishment and Molecular Profiling of Lung Squamous Cell Carcinoma Patient-derived Xenograft Models.

Purpose : Lung squamous cell carcinoma(SqCC) has limited targeted therapeutic options in comparison with adenocarcinoma(ADC). Patient-derived xenograft(PDX) is an effective tool to preserve or study the human tumors in vivo. We established the PDX models of primary lung SqCC PDX models to be used for the discovery of target mutations through the molecular profiling.

Methods : PDX models were established using the tumor tissues of patients who underwent surgery as primary lung SqCC at Samsung Medical Center during the period between November, 2014 and January, 2019. The tumor tissues from patients were subcutaneously engrafted and passaged two more times in NOD-scid-IL2Rγnull mouse. Then histological characteristics and molecular profiles of established SqCC PDX were analyzed with whole exome sequencing and whole transcriptome sequencing.

Results : Total 139 lung SqCC samples were enrolled and implanted in NSG mice to generate PDX model. 82 cases generated PDX engraftment and 57 cases were no-growth. Among 82 PDX engraftments, 62 cases successfully generated SqCC PDXs (Success rate = 43.66%) and 20 cases included xenograft-associated lymphoproliferative diseases and others. The growth speed of PDX engraftment was around 4 months (average 12.5 weeks, 3.2 to 34.7 weeks). Successful PDX engraftment slightly correlated with overall survival (p=0.023, hazard ratio [HR]=2.36, 95% confidence interval [CI] = 1.13 to 4.94). Among them, some targetable mutations including BRAF(V600E) were identified.

Conclusion : The PDX models have been successfully established from the human tumors of the primary lung SqCC. We revealed that PDX models have closely concordance with the original human tumor by comparing the pathology data and molecular profiling.
