Overexpression of Collagen Triple Helix Containing-1 in Esophageal Squamous Cell Carcinoma : Functional Analysis and Clinical Significance

목적 : The prognosis of patients with esophageal squamous cell carcinoma (ESCC) still remains poor, despite of the resent advancement of therapeutic modalities. So, we have screened new biomarkers of ESCC by genomic profiling and validation process. After this validation process, we found that Collagen Triple Helix Containing-1(CTHRC1) was significantly overexpressed in ESCC and potentially correlated with proliferation and prognosis. In this study, we evaluated function and clinical significance of this potential biomarker.
방법 : We performed RT-PCR, western blot, and immunohistochemical staining (IHCS) to verify the expression pattern and the prognostic implications of CTHRC1 in ESCC. Tissue array method was used for IHCS of paraffin blocks of 157 surgical specimen. Functional analysis was conducted by comparing the function of TE-8 and CTHRC1 knock-down TE-8 ESCC cell line. A wound healing assay, invasion assay and colony formation assay were performed.
결과 : Comparison of genome expression data between ESCC and normal tissues of esophageal mucosa revealed a significant increase (~24.4-fold) in CTHRC1 transcripts in cancer tissues samples. Both RT-PCR and Western blot analysis revealed that CTHRC1 was significantly upregulated in tumor tissue samples of ESCC. The inhibition of CTHRC1 expression showed significantly decreased proliferation, migration, invasion, and colony formation in ESCC TE-8 cell line. The percentage of positively stained cells by IHCS staining showed inversed correlation with overall survival (p=0.035).
결론 : Overexpression of CTHRC1 was observed quite frequently in ESCC tissues and cell lines. In this study, we first found that CTHRC1 was associated with cell proliferation, migration, invasion, and colony forming ability in ESCC. The higher population of CTHRC1 positive ESCC cells was correlated with the worse prognosis. Although further study for CTHRC1 will be required to identify its molecular regulation, it might be one of the useful molecular biomarkers for ESCC.
